CD137 protein is fully biologically active when compared to standard. The biological activity is determined by its inhibitory effect of IL-8 production using human peripheral blood mononuclear cells. About 90 % of inibition was seen using a concentration of 1 …
CD137, a member of the tumor necrosis factor receptor superfamily, represents a promising target for enhancing antitumor immune responses (1). The functions of CD137 in T lymphocytes include regulating activation, proliferation and apoptosis (2). CD137 helps regulate the activation of many immune cells, including CD4(+) T cells, CD8(+) T cells, dendritic cells, and natural killer cells. CD137 is a 39 kD transmembrane protein also known as 4-1BB. It is expressed on activated T cells.
CD137-transfected L-428 cells (L-428-CD137) were treated with 10 μg/ml of Fabs, followed by anti-Fab-AF647. All Fabs bound efficiently to CD137 + cells ( Supplementary Figure 3 ). No binding was observed to CD137 − L-428-control cells (not shown). 137A and 137C showed weaker binding affinity than the other Fabs, making them suitable for the generation of the bispecific antibody.
CD137, also known as TNFRSF9 or 4-1BB, is an inducible costimulatory molecule expressed mainly on activated T cells. Its ligand, known as 4-1BBL, is expressed on activated macrophages, mature B cells, hematopoietic stem cells, and myeloid progenitor cells.
137A and 137C showed weaker binding affinity than the other Fabs, making them suitable for the generation of the bispecific antibody. The CD137 receptor (4-1BB, TNF RSF9) is an activation induced molecule expressed by antigen-specific T-cells.
CD137 (4-1BB, Tnsfr9) is a member of the TNF-receptor (TNFR) superfamily without known intrinsic enzymatic activity in its cytoplasmic domain. Hence, akin to other members of the TNFR family, it relies on the TNFR-Associated-Factor (TRAF) family of adaptor proteins to build the CD137 signalosome for transducing signals into the cell.
Methods Foxp3+ and CD8+ T cells in GCs were investigated using immunohistochemistry (IHC). CD137 expression in GCs was detected using flow cytometry, IHC and immunofluorescence (IF). Peripheral blood mononuclear cells (PBMCs 2019-11-08 · The tumor necrosis factor receptor superfamily member 9 (TNFRSF9), also known as CD137 or 4-1BB, is another important immune-modulating molecule known to be expressed on the surface of certain immune cells. CD137 was originally discovered in 1989 and reported as a cell surface protein mainly located on activated T cells .
TNFRSF9 / 4-1BB / CD137 is a member of the TNF-receptor superfamily. This receptor contributes to the clonal expansion, survival, and development of T cells. It can also induce proliferation in peripheral monocytes, enhance T cell apoptosis induced by TCR/CD3 triggered activation, and regulate CD28 co-stimulation to promote Th1 cell responses. CD137 ligand signaling also enhances monocyte migration in vitro and in vivo. Immobilized rCD137 protein induced the movement of peripheral human monocytes in vitro [26]. Fur-ther, 6 times as many monocytes infiltrated CD137-express-ing cell spheroids than control spheroids.
